A little surprised at no mention of Daniel Plainview yet. (Even though I feel like that movie is a tad overrated.)
That said, I had no idea that movie was loosely based on a novel, even though the character had a different name.

William Costigan Jr in The Departed. Great character. Loses weight throughout the film due to stress and has a pain pill addiction. Despite all the stress he still manages to infiltrate the gang. One of my favorite films.
I forgot his name was Hans Lands but just looked up Inglorious *******s and that's exactly who I would say fits this thread.
At least honorable mention for Cliff Booth-Once Upon a Time in Hollywood
