To see this for the first time, with the dramatic John Williams score, the sheer size of the ship filling the entire screen and the sound of the laser fire back and forth...........it was awe-inspiring and can't be matched.

Lots of good ones already mentioned, so I'll throw the dairy farm scene from "Inglourious Basterds." May be Tarantino's finest moment.

I would say the same about Halloween and what it did for the horror/scary movie genre although I wouldn't necessarily say it's the best opening scene. I would agree that the above is.GiveEmHellBill said:

The opening scene of La La Land is a continuous shot choreographed song and dance number while shutting down a freeway. I'm not big into musicals, but from a cinematography standpoint, that's pretty awesome.
