I could list many, but I do recall my first time. I saw Airplane when I was 11, and I was cry/laughing a handful of times throughout that movie.
There're two for me as well:
First is kinda lame but it's the Pixar animated short "Lifted." Cannot sit through it without having to pause it.

Don't know how to post YouTube videos, but the scene that comes to mind is from Harlem Nights. The part where they are asking the champ if he is ready for the big fight, and he stutters the whole scene.

The Bridesmaid diarrhea scene has been mentioned, but that made me have tears. Also, the one scene in Hangover 2 when they were retracing their steps and it comes out the Stuart slept with a ****** and he was describing how they both came together and how beautiful it was. Alan realizes there were chicks with d***s, and he says, "what is this a magic show?", I lost it. I would post the scene, but I would be banned.
