I would second Capetown. We were there last year, although we stayed in Stellenbosch (check out Sugarbird Manor). Had a great time - went wine tasting, checked out some beaches, hit South Cape and had a great evening at the V&A. Long flight, but combine it with some other activities and it is so worth it.

3rd for Cape Town. This incredibly small area is home to its own plant and animal kingdoms.... life you don't see elsewhere on Earth. Plus Table Mountain is one of a kind. Epic.

When you get to 50yrs old you spend more and more of your own dime to check mark the bucket listers: Booked for Antarctic cruise next December for the Total Solar Eclipse. Goal is to touch 'em all!

All that being said, Greek Islands - fantastic place to spend someone else's money.

Antarctica would be my initial thought. That or the most epic European ski trip imaginable
