Twitter is good for some things -

I'd go with
Jimmy Chitwood shooting the winning shot in Hoosiers
Andy Defrain escaping in Shawshank
Red arriving in Zihuatenejo also in Shawshank
Courtroom scene in A Few Good Men
Phoebe Cates in the pool Fast Times at Ridgemont High

I always thought this Hoosiers scene was kind of subtly amazing and fantastic. Not necessarily "better" than the big game winning shot but just done so well.

Jimmy is alone shooting out side on a dirt court and coach comes up to talk to him - and he never misses the entire time he is shooting and just listens as coach talks. When coach says he doesn't care if he plays or not and walks off jimmy finally misses.

It rattled him. and it was just perfect.

I think my absolute favorite though and I think a perfect scene is the park bench scene in Good Will Hunting.

It comes after the previous session when Will had gutted Sean (Robin Williams) in their previous meeting analyzing him from a single painting and hitting him where it hurts - his dead wife.

Starts off with more smartass stuff from will but Sean soon starts a long monologue here where he methodically tears Will apart not in an angry way but destroying his entire self image really. the camera work is amazing panning around where first you only see Sean and then you get Wills stunned blank stare as he is just rocked by this and taking it in. He rips his attitude and demeanor apart but in a way that shows he wants to help him still. And in the end he gives him a "your move chief" and walks off as Will sits stunned still taking it in.

Perfection

Although I'll nominate another - the Godfather Baptism scene is brilliant.

the juxtaposition of the somber and sacred baptism and Michael denouncing satan and sin, intermixed with the various hits as he eliminates his enemies is perfection.

Yup. Was just about to post this. I've seen Jaws hundreds of times it seems like, and this scene never fails to give me goosebumps. As technically great a filmmaker Spielberg is, it's pretty crazy that this scene is probably the best one he ever shot. It's a couple of cameras, an actor doing his best work, and the sound of whales in the background, and it's absolutely captivating. I've always said that if they-god forbid-ever remake Jaws, lesser filmmakers would resort to filming some elaborate flashback of the Indianapolis that would undoubtedly cost millions and have exactly zero the impact that this scene does.

Immediately thought of 2 baptism scenes. Since the first one was already posted, I'll go with the other one:

Excellent additions so far. I'll add two of what I think are the most powerful scenes from one of my all-time favorite movies, Amadeus. The first is Salieri's coming to grips with Mozart's genius. I love how well the scene is composed switching from piece to piece and watching him become more enraged...lost in the music and spilling it all on the ground. I wished this clip wasn't cut right then because the scene ends with him admitting that it's "Miraculous".

The 20 second section from 1:20 to 1:40 gives me chills every time! What he is saying, the music, and the priest's response is absolute perfection. I LOVE this scene!

The ease with which he lies to her, and the momentary relief she feels, only to realize quickly that she had been lied to, that he will be no different than his father. So perfect.

yeah so good and i go back further than that clip although it is not "one scene" but from the point that he leaves them at the waterfall where they are separated and even the entire indian villlage scene with Nathaniel coming in unarmed it all plays out on an incredible epic but personal scale.

the fear of the sisters being separated and all the negotiations
Duncan's noble swap of the girls for himself (the translation parts between Huron, French and English handled so well)
the mercy killing of Duncan
Uncas frantic chase after them through the mountains one against many
until the gut wrenching death of Uncas
the desperation of the sister Alice to jump from the cliffs
Magua's indifference and lack of comprehension
Cora's silent but painful reaction
the father Chingachgook frantic chase and Nathaniel having his back the entire way
the death of Magua at the father's hands

and all of it against an incredible scenic backdrop and with that epic score

the burial of Uncas is serenely beautiful and the reveal of Chingachgook as the titular last of the mohicans (if you had not read the book) was poetically perfect and sad.
