New York City for the somewhat experienced tourist

My wife and I are headed to NYC for a long weekend the last weekend of this month to celebrate our 5-year wedding anniversary. We have been twice before and have done WTC, Top of the Rock, Ferry tour, and some Central Park.

My question is what should we do next? We are staying at the Loews Regency (61st and Park) so easy access to most places. The High Line is on my list this time, as is Brooklyn (but not exactly sure what to do there). Any advice is appreciated.
